Tirzepatide (M-ounjaro) is a novel medication indicated as an adjunct to diet and exercise in the treatment of patients with type 2 diabetes mellitus. Tirzepatide is a dual receptor agonist, acting on glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) to lower blood glucose levels.
Tirzepatide is a dual glucose-dependent insulinotropic polypeptide (GIP) and GLP-1 agonist that has been studied recently as a treatment for patients with noncirrhotic NASH (SYNERGY-NASH, NCT04166773) given its association with significant weight loss and improvement in features of metabolic syndrome in diabetes trials.
Tirzepatide is used to improve glycaemic control in adults with type 2 diabetes. It is the first glucose-dependent dual agonist of glucagonotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. According to its SURPASS series of studies, the highest dose of 15mg further reduces HbA1C by up to 1.6% absolute compared to placebo. In addition to its glucose-lowering effects, the SURMOUNT-1 study in obese patients without diabetes showed that the highest dose of 15 mg of Tirzepatide significantly reduced patient body weight by an average of 22.5% (24 kg).
